Abstract Cadmium (Cd) is one of the most toxic elements to plants, so Cd-tolerant trees should be used in reforestation or remediation systems. In this study the effects of Cd on seed germination and early seedling growth were evaluated in Schinus molle plants. Seeds of S. molle were treated with Cd at concentrations of 0 (control), 0.01, 0.02, 0.05, and 0.125 mM using Cd(NO3)2. Seed germination parameters and seedling growth traits were evaluated 33 days after the experiment started. Seeds and roots were sampled for anatomical evaluation and processed with plant microtechniques. No effects of Cd were found on the germination percentage of S. molle seeds. However, seedlings’ shoot and root fresh masses and lengths increased with Cd treatment. Cadmium increased root cortex and endodermis thickness and reduced the percentage of vascular tissues in seedlings. Schinus molle seeds and seedlings thus showed tolerance to Cd related to anatomical adaptations.

Em razão do prognóstico de mudanças climáticas nos próximos anos, a compreensão do déficit hídrico e das respostas fisiológicas de plantas ao mesmo, torna-se uma importante condição para o desenvolvimento de tecnologias, como mecanismos que auxiliem as plantas a suportar períodos mais longos de seca, o que será essencial para a manutenção da produção mundial e brasileira. Neste estudo, objetivou-se avaliar aspectos ecofisiológicos e anatômicos além da atividade da redutase do nitrato em mudas de café Siriema submetidas a quatro tratamentos: irrigadas diariamente, não irrigadas, re-irrigadas 24 horas e re-irrigadas 48 horas após os diferentes períodos de estresse. A ausência de irrigação promoveu redução no potencial hídrico foliar, sendo mais acentuada a partir do nono dia de avaliação. A re-irrigação das mudas promoveu uma recuperação parcial do estado hídrico das plantas. Nas plantas não irrigadas observou-se um aumento na resistência estomatal e diminuição da transpiração, reduzindo a atividade da redutase do nitrato nas folhas. Nas raízes, houve um declínio na redutase do nitrato sob estresse hídrico. Ocorreram modificações na anatomia foliar apenas na espessura da epiderme da face adaxial e parenquimama paliçádico, sendo maior nas plantas controle. A densidade estomatal e razão diâmetro polar e equatorial demonstraram maiores valores nas plantas sob déficit hídrico. Nas raízes, foi observada diferença apenas na espessura do córtex, sendo esta maior no tratamento não irrigado. Dessa forma, o café Siriema sob condições de estresse hídrico apresenta modificações fisiológicas, bioquímicas e anatômicas que contribuem para a tolerância desse genótipo nessas condições.

Due to the weather changes prognostic for the coming years, the understanding of water deficit and physiological responses of plants to drought becomes an important requirement in order to develop technologies such as mechanisms to assist plants to cope with longer drought periods, which will be essential to maintenance of Brazilian and worldwide production. This study aimed to evaluate ecophysiological and anatomical aspects as well as the nitrate reductase activity in Siriema coffee seedlings subjected to four treatments: Daily irrigated, non-irrigated, re-irrigated 24 hours and re-irrigated 48 hours after different stress periods. Non-irrigation promoted a reduction in leaf water potential being accented from the ninth day of evaluation onwards. Re-irrigation promoted a partial recovery of the plant water potential. Non-irrigated plants showed an increase in stomatal resistance and reduction of transpiration and nitrate reductase activity. In the roots, there was a decrease in nitrate reductase activity under water stress. Leaf anatomical modifications were significant only for the adaxial surface epidermis and palisade parenchyma thickness, this latter characteristic being higher in control plants. Stomatal density and polar and equatorial diameter ratios showed the highest values in plants under water stress. In the roots, differences only in the cortex thickness being bigger in the non-irrigated treatment could be observed. Therefore, Siriema coffee plants under water stress show physiological, biochemical and anatomical modifications that contribute to the tolerance of this genotype to these conditions.

A cultura do café é de expressiva importância no mercado internacional, com uma produção anual de cerca de 120 milhões de sacas beneficiadas e no Brasil, é responsável pela geração de mais de dez milhões de empregos diretos e indiretos. O estresse hídrico é um fator ambiental que pode ser extremamente prejudicial para a cultura do café, sendo que a seleção de cultivares tolerantes é de grande importância, principalmente diante da expansão da cafeicultura brasileira para áreas consideradas marginais para o cultivo do café por estarem sujeitos à deficiência hídrica. Nesse trabalho foram avaliadas 15 cultivares de Coffea arabica quanto ao status hídrico e às características da estrutura interna das folhas, verificando quais dessas cultivares são potencialmente mais eficientes em tais condições. Dentre as cultivares estudadas destacam-se a Bourbon Amarelo e a Catimor como potencialmente mais eficientes em condições de estresse hídrico por possuírem: maior potencial hídrico, cutícula mais espessa, maior proporção de parênquima paliçádico, maior espessura da nervura central e maior densidade estomática em relação às demais variáveis. Dessa forma, as cultivares Bourbon Amarelo e Catimor são potencialmente mais eficientes na redução da transpiração, fotossíntese e translocação de fotossintatos em condições de estresse hídrico, sendo mais indicadas ao plantio em regiões sujeitas a maiores déficits hídricos em relação às demais cultivares estudadas.

Coffee yield has a great importance to the international market with a 120 million 60-kilogram bags and in Brazil it produces 10 million of direct and indirect jobs. Water stress is one of the most important factors to the crop yield, and so, the selection of tolerant cultivars is of great importance in the places under water stress. In this work, 15 Coffea arabica cultivars were evaluated about the water status and leaf anatomy to indicate the most tolerant Cultivars under water stress. The Bourbon Amarelo and Catimor were the most efficient under water stress, because these varieties have thicker cuticle, more palicade parenchyma, bigger central bundle sheath, and higher stomatal density. So, these Cultivars can be more potentially more efficient to reduce the transpiratory rate, to improve the photosynthesis and photosynthates translocation under water stress, and these varieties can be more indicated under water stress against the other cultivars.

O objetivo deste trabalho foi avaliar o efeito de sucessivos ciclos de seleção na plasticidade foliar do milho 'Saracura' BRS-4154 sob alagamento intermitente do solo em condições de campo. O alagamento do solo iniciou-se no estádio de seis folhas com a aplicação de uma lâmina de 20 cm de água três vezes por semana. No florescimento, foram coletadas e fixadas amostras das folhas. Secções paradérmicas e transversais de amostras foram observadas em microscópio fotônico. Várias modificações foram observadas durante os ciclos de seleção, como o número e o tamanho dos estômatos, a maior quantidade de feixes vasculares e a consequente diminuição da distância entre eles, o metaxilema com diâmetros menores, a diminuição da espessura da cutícula e da epiderme, a diminuição no número e tamanho das células buliformes, o aumento da espessura do floema e a menor área de esclerênquima. Os sucessivos ciclos de seleção do milho 'Saracura', portanto, levaram a mudanças na anatomia foliar, as quais podem ser favoráveis à tolerância da planta ao alagamento intermitente do solo.

The objective of this work was to assess the effect of successive selection cycles on leaf plasticity of 'Saracura' maize BRS-4154 under periodical flooding in field conditions. Soil flooding started at the six-leaf stage with the application of a 20-cm depth water layer three times a week. At flowering, samples of leaves were collected and fixed. Paradermic and transverse sections were observed under photonic microscope. Several changes were observed throughout the selection cycles, such as modifications in the number and size of the stomata, higher amount of vascular bundles and the resulting decrease of the distance between them, smaller diameter of the metaxylem, decrease of cuticle and epidermis thickness, decrease of number and size of bulliform cells, increase of phloem thickness, smaller sclerenchyma area. Therefore, the successive selection cycles of 'Saracura' maize resulted in changes in the leaf anatomy, which might be favorable to the plant's tolerance to the intermittent flooding of the soil.

Compara-se neste trabalho a distribuição de genótipos do sistema sangüíneo Duffy, determinados através de reação em cadeia da polimerase com oligonucleotídeos iniciadores alelo-específicos, em 68 pacientes com infecção por Plasmodium vivax e em 59 controles com malária não-vivax de Rondônia, Brasil. Nenhum paciente infectado com P. vivax, mas 12% dos controles não-vivax, eram homozigotos para o alelo Fy, que abole a expressão do antígeno Duffy em hemácias. No entanto, não se observou evidência de proteção significativa contra P. vivax entre indivíduos heterozigotos para Fy. O alelo Fy x, que tem sido recentemente associado com a expressão eritrocitária muito fraca do antígeno Duffy, não foi encontrado entre pacientes locais com infecção por P. vivax.

We have compared Duffy blood group genotype distribution, as determined by polymerase chain reaction with allele-specific primers, in 68 Plasmodium vivax-infected patients and 59 non-vivax malaria controls from Rondônia, Brazil. Homozygosity for the allele Fy, which abolishes Duffy antigen expression on erythrocytes, was observed in 12% non-vivax controls but in no P. vivax patient. However, no significant association was found between Fy heterozygosity and protection against P. vivax. The Fy x allele, which has recently been associated with very weak erythrocyte expression of Duffy antigen, was not found in local P. vivax patients.

Dois protocolos de reação em cadeia da polimerase (PCR) apresentaram baixa sensibilidade (36% e 53%, respectivamente), para TB AMPLICOR e PCR aninhado baseado no gene MPB64), quando comparados aos métodos microbiológicos clássicos (73% e 54% respectivamente para baciloscopia e cultura), no diagnóstico de meningite tuberculosa em 91 pacientes do sudeste do Brasil. Somente três pacientes apresentaram PCR positiva e microbiologia negativa. A análise de amostras seqüenciais de líquor com a PCR aninhada detectou DNA de Mycobacterium tuberculosis até 29 dias após a introdução de tratamento.

Two polymerase chain reaction (PCR) protocols showed low sensitivity (36% and 53% for TB AMPLICOR and MPB64 nested PCR, respectively), when compared with classic microbiological methods (73% and 54% for Ziehl-Neelsen staining and culture, respectively), in the diagnosis of tuberculous meningitis in 91 patients in southeastern Brazil. Only three PCR-positive, microbiologically negative patients were found. Analysis of sequential cerebrospinal fluid samples by nested PCR detected Mycobacterium tuberculosis DNA up to 29 days after the introduction of antituberculosis chemotherapy.
